应用螯虾血淋巴细胞的原代培养系统 ,作为对虾白斑杆状病毒 (whitespotbacilliformvirus ,WSBV )的替代宿主 ,进行体外感染实验。通过PCR及RNA点杂交的方法监测WSBV的DNA复制、转录情况。结果显示 ,来源于对虾的WSBV可以感染体外培养的螯虾原代血淋巴细胞 ,并在其中增殖 ;WSBV感染螯虾原代血淋巴细胞后 6h左右病毒开始大量复制 ,感染后的细胞可以存活数日。在目前虾细胞系尚未建立之前 ,螯虾血淋巴原代细胞可以作为研究WSBV感染、转录和复制等机理的替代系统。
In vitro infection experiments were performed using a primary culture system of crayfish hemolymph cells as a surrogate host for whitespotbacilliformvirus (WSBV). The DNA replication and transcription of WSBV were monitored by PCR and RNA dot hybridization. The results showed that WSBV derived from shrimp could infect and proliferate primary hemocytes of crayfish cultured in vitro. After 6 hours of primary hemolymph cells from WSBV infected with crayfish, the virus began to replicate and the number of surviving cells after infection day. Prior to the current shrimp cell line has not been established, the hemopterobrotimal hemolymph primary cells can be used as an alternative system to study WSBV infection, transcription and replication mechanisms.
