【目的】探讨补肾化瘀法抗光老化对基质金属蛋白酶MMP-1与MMP-3表达的影响。【方法】采用8-甲氧沙林(8-MOP)/长波紫外线照射SD大鼠法,复制光老化大鼠模型,并对该动物模型给予补肾化瘀中药干预,连续15 d,采用免疫组织化学SP法,分别检测空白对照组、补肾化瘀中药干预组和光老化模型组大鼠皮肤MMP-1、MMP-3的表达,使用图像分析软件分析各组照片的单位灰度值并进行比较,根据灰度值的大小判断MMP-1、MMP-3的表达水平。【结果】建立了光老化大鼠模型。经过免疫组织化学染色显示,补肾化瘀中药干预组大鼠皮肤切片中阳性染色颗粒明显减少,MMP-1和MMP-3表达显著减弱。使用图像分析软件分析结果显示,光老化模型组大鼠皮肤中MMP-1和MMP-3表达显著高于空白对照组,差异有统计学意义(P<0.05);中药干预组大鼠皮肤中MMP-1和MMP-3表达水平显著降低,与模型对照组比较,差异有统计学意义(P<0.05)。【结论】补肾化瘀中药能显著降低光老化模型大鼠皮肤中MMP-1和MMP-3的表达水平,促进光老化大鼠皮肤损害的修复,具有一定的抗光老化作用。
【Objective】 To investigate the effects of the method of tonifying kidney and removing blood stasis on the expressions of matrix metalloproteinase-1 and matrix metalloproteinase-3 (MMP-3). 【Method】 The photo-aging rat model was reproduced by 8-methoxsalin (8-MOP) / ultraviolet (UV) irradiated SD rats. The animal model was given TCM intervention of tonifying kidney and removing blood stasis for 15 days. The chemical SP method was used to detect the expression of MMP-1 and MMP-3 in the skin of rats in the control group, Bushen Huayu TCM intervention group and the photo-aging model group respectively. The gray value of each unit was analyzed by image analysis software, According to the size of the gray value to determine the expression of MMP-1, MMP-3. 【Result】 A photoaging rat model was established. Immunohistochemical staining showed that the number of positive staining particles in the skin sections of rats in the Bushen Huayu intervention group was significantly decreased, and the expressions of MMP-1 and MMP-3 were significantly weakened. The results of image analysis software showed that the expression of MMP-1 and MMP-3 in the skin of the photoaging group was significantly higher than that of the blank control group (P <0.05) 1 and MMP-3 expression levels were significantly lower than the model control group, the difference was statistically significant (P <0.05). 【Conclusion】 Bushen Huayu herbs can significantly reduce the expression of MMP-1 and MMP-3 in the skin of photo-aged rats and promote the repair of skin damage in photo-aged rats, and have certain anti-photoaging effect.
