目的:建立正常人胆囊组织基因表达目录。方法:采用含17000cDNA克隆的基因微矩阵与2例正常胆囊之cDNA杂交,取基因表达平均值,分析胆囊的基因表达谱。结果:正常胆囊有11047条基因表达,其前10位高表达基因与平滑肌收缩及物质转运有关。在前200位高表达的基因中,离子通道和转运蛋白、蛋白翻译和合成类以及发育相关类基因占多数。在胆囊所有表达的基因中,12、2、21号染色体表达的数目最多。另外,21条胆石病候选基因在胆囊中有表达,其中主要为脂类代谢相关基因。两条胆囊特异性表达基因:NPC1L1、SLC17A2。结论:胆囊高表达的基因可能与胆囊收缩和胆汁浓缩有关,胆囊不仅参与机体的消化功能,还参与调节脂类代谢平衡。
Objective: To establish a catalog of gene expression in normal human gallbladder. Methods: A cDNA microarray containing 17000cDNA clones was used to hybridize with two normal gallbladder cDNAs. The mean gene expression was analyzed and the gallbladder gene expression profile was analyzed. Results: There were 11047 genes in normal gallbladder. The top 10 genes were related to the contraction and substance transport of smooth muscle. Among the top 200 highly expressed genes, ion channels and transporters, protein translation and synthesis, and developmental related genes are the majority. Of all the genes expressed in the gallbladder, chromosomes 12, 2 and 21 have the highest number of expressed genes. In addition, 21 gallstone candidate genes are expressed in the gallbladder, mainly lipid metabolism related genes. Two gallbladder specific expression genes: NPC1L1, SLC17A2. Conclusion: The high expression of gallbladder genes may be associated with gallbladder contraction and bile concentration related to the gallbladder not only involved in the body’s digestive function, but also involved in the regulation of lipid metabolism.
