对目前文献所见到的植酸酶活的测定方法选取三种进行了分析和修正,并用两种温度下的酶活定义检验修正过的测定方法的差异。根据显色剂吸收光谱和磷化合物的吸收光谱的比较,确定FeSO_4—钼蓝法选取大于720nm、VC—钼蓝法选取620~660nm、丙酮法选取400~420nm的波长为测量适宜波长。在同样温度下(即对一种酶活定义来说,)用这三种方法测定曲霉发酵液的植酸酶活,结果酶活大小基本一致(平均差率小于5%),说明这三种方法在测定植酸酶活时都有相等的可靠性和准确性。
Three methods were selected for the determination of phytase activity as seen in the current literature, and the differences between the modified assays were tested using enzyme definitions at two temperatures. According to the comparison between the absorption spectrum of the chromogenic reagent and the absorption spectrum of the phosphorus compound, it is determined that the wavelength selected by the FeSO_4-molybdenum blue method is greater than 720 nm, the wavelength selected by the VC-molybdenum blue method is 620~660 nm, and the acetone method selected by the wavelength of 400~420 nm is the suitable wavelength for measurement. The phytase activity of the Aspergillus fermentation broth was determined by the three methods at the same temperature (ie for the definition of an enzyme activity), with the result that the size of the enzyme activity was essentially the same (mean difference less than 5%), indicating that the three The method has equal reliability and accuracy in determining phytase activity.
