目的应用U937泡沫细胞模型建立一种新的检测高密度脂蛋白(HDL)外运细胞内胆固醇生物学活性的方法。方法将U937细胞与100μg/mL氧化低密度脂蛋白孵育36h后,分别用不同浓度高密度脂蛋白处理24h,应用高效液相色谱(HPLC)检测细胞内胆固醇和胆固醇酯含量。结果经氧化低密度脂蛋白处理后的U937细胞,HPLC检测细胞内总胆固醇增加,胆固醇酯含量大于胆固醇含量;经高密度脂蛋白处理后,细胞内胆固醇含量减少,不同厂家及不同批次的高密度脂蛋白外运胆固醇的生物学活性有所不同。结论经氧化低密度脂蛋白处理后的U937细胞可以作为检测高密度脂蛋白外运胆固醇生物学活性的方法。
Objective To establish a new method for detecting the intracellular cholesterol bioactivity of high density lipoprotein (HDL) exocytosis cells by U937 foam cell model. Methods U937 cells were incubated with 100μg / mL oxidized low density lipoprotein for 36h. The cells were treated with different concentrations of high density lipoprotein for 24 hours respectively. The contents of intracellular cholesterol and cholesteryl ester were determined by high performance liquid chromatography (HPLC). Results The U937 cells treated with oxidized low density lipoprotein were assayed for the increase of intracellular total cholesterol and cholesterol ester content by HPLC. After high density lipoprotein treatment, the intracellular cholesterol content decreased, The biological activity of LDL cholesterol is different. Conclusions U937 cells treated with oxidized low-density lipoprotein can be used as a method to detect the biological activity of high-density lipoprotein cholesterol.
