目的研究运动大鼠骨骼肌细胞线粒体膜电位的变化与细胞凋亡的关系。方法用流式细胞仪 (flowcytometry ,FCM)检测肌细胞线粒体膜电位、DNA倍体分析及脱氧核糖核苷酸末端转移酶介导的缺口末端标记技术 (terminaldeoxynucleotidyltransferasemediatedd UTPnickendlabeling ,TUNEL)。 结果线粒体膜电位在游泳运动后第 1周降低 ,第 2周后开始明显增高 ,第 3周略有降低 ,运动后各组的DNA均出现亚“G1”峰即凋亡峰。TUNEL显示 ,运动后第 1周凋亡比例最高 ,第 2周后比例降低。结论运动引起线粒体膜电位的改变 ,该方法快速、特异性强、灵敏度高
Objective To investigate the relationship between mitochondrial membrane potential changes and apoptosis in skeletal muscle cells of exercise rats. Methods Flow cytometry (FCM) was used to detect mitochondrial membrane potential, DNA ploidy analysis and terminal deoxynucleotidyl transferase mediated nick end labeling (TUNEL). Results The mitochondrial membrane potential decreased in the first week after swimming, then increased significantly after the second week and decreased slightly in the third week. After the exercise, the mitotic membrane sub-G1 peak or apoptotic peak appeared. TUNEL showed the highest proportion of apoptotic cells in the first week after exercise, and decreased after the second week. Conclusion Exercise induced mitochondrial membrane potential changes, the method is rapid, specific and sensitive
