目的探讨新型Rho激酶抑制剂FSD-C10对A53T细胞突起的影响及其促进突起生长的机制。方法 A53T细胞进行常规培养和传代,细胞生长到90%左右后,根据不同实验的需要,分为PBS阴性对照组、Fasudil阳性对照组及FSD-C10干预组。应用Image-Pro Plus v6.0软件测量不同时间各组细胞突起的长度,Western blot检测突起相关蛋白synapsinⅠ、synaptophysin、PSD-95的表达情况。结果 FSD-C10促进A53T细胞突起的生长,synapsinⅠ蛋白表达升高。结论 FSD-C10可能通过诱导synapsinⅠ的表达实现对突起生长的促进作用。
Objective To investigate the effects of a novel Rho kinase inhibitor FSD-C10 on the prominence of A53T cells and its mechanism of promoting neurite outgrowth. Methods After routine culture and passage of A53T cells, the cells were divided into PBS-negative control group, Fasudil-positive control group and FSD-C10-treated group according to the needs of different experiments. Image-Pro Plus v6.0 software was used to measure the length of the cell protrusion in each group at different time points. Western blot was used to detect the expression of synapsin I, synaptophysin and PSD-95. Results FSD-C10 promoted the growth of A53T cells and increased the expression of synapsin I protein. Conclusion FSD-C10 may promote the process of neurite outgrowth by inducing synapsin Ⅰ expression.
