目的:观察不同剂量~(131)Ⅰ-HAb18在长期毒性试验中对大鼠各组织的病理损伤,为确定安全的临床使用剂量提供依据。方法:80只SD大鼠随机分为4组(含对照组),实验组腹腔注射3种剂量的~(131)Ⅰ-HAb18,对照组注射等体积生理盐水,每月2次,共3月,末次注射24h后处死,解剖一半大鼠,取各组织制成石蜡切片,镜检。停药恢复2月后,处死解剖余下的大鼠,制成切片,观察。结果:与对照组相比,低剂量组的大鼠各组织基本正常,中剂量组的大鼠约有一半组织出现轻度的病理改变,高剂量组的大鼠部分组织出现坏死等不可复性病理改变。结论:在长期毒性试验中低剂量为安全剂量,中剂量对SD大鼠有毒性作用,高剂量对SD大鼠有较重的毒性。
OBJECTIVE: To observe the pathological damage of rat 131I-HAb18 in different doses of long-term toxicity test in order to provide evidence for the safe clinical dosage. Methods: Eighty SD rats were randomly divided into 4 groups (control group). The experimental group was injected intraperitoneally with 131I-HAb18 131I, and the control group was injected with equal volume of normal saline twice a month for 3 months , 24 h after the last injection was sacrificed, dissected half of the rats, and made of paraffin sections of tissue, microscopic examination. Two months after discontinuation of treatment, the remaining rats were sacrificed and sliced to observe. Results: Compared with the control group, the rats in the low-dose group were basically normal, about half of the rats in the middle-dose group had mild pathological changes, and the necrosis of some tissues in the high-dose group was irreversible Pathological changes. Conclusion: In the long-term toxicity test, low dose is a safe dose, middle dose is toxic to SD rats and high dose to SD rats is more toxic.
