目的探讨吗啡预处理对兔缺氧复氧损伤肾脏组织的超氧化物歧化酶(SOD)和丙二醛(MDA)的影响。方法雄性新西兰大白兔27只随机分成正常对照组(N组)、缺氧复氧组(H/R组)和吗啡预处理组(MO+H/R组)。H/R组和MO+H/R组分别静脉注射生理盐水5 mL和吗啡3 mg/kg后吸入8%O23 h后空气复氧3h。比较各组肾脏组织SOD、MDA水平和肾脏含水量。结果与N组比,H/R组明显降低SOD活力〔(226±13.3)U/g,P<0.05〕,增加MDA含量〔(29.02±2.15)U/g,P<0.05〕。与H/R组相比,吗啡缺氧预处理明显增加了肾脏组织SOD活力〔(294±14.1)nmol/L,P<0.05〕,降低肾脏组织MDA含量〔(16.55±2.23)nmol/L,P<0.05〕。3组兔肾组织水含量差异无统计学意义。结论吗啡缺氧预处理可能减轻缺氧复氧损伤肾脏中MDA的生成,增强SOD的活力,从而发挥对兔缺氧复氧损伤肾脏的保护作用。
Objective To investigate the effects of morphine preconditioning on superoxide dismutase (SOD) and malondialdehyde (MDA) in kidney of hypoxia-reoxygenation injury in rabbits. Methods Twenty - seven male New Zealand white rabbits were randomly divided into normal control group (N group), hypoxia - reoxygenation group (H / R group) and morphine preconditioning group (MO + H / R group). In the H / R group and the MO + H / R group, 5 mL of normal saline and 3 mg / kg of morphine were respectively inhaled for 8 h and then reoxygenated for 3 h. The levels of SOD and MDA and the water content of kidneys in each group were compared. Results Compared with group N, the activity of SOD in H / R group was significantly lower than that in group N (226 ± 13.3 U / g, P <0.05), and MDA content was increased (29.02 ± 2.15 U / g, P <0.05). Compared with H / R group, morphine hypoxic preconditioning significantly increased the activity of SOD in renal tissue 〔(294 ± 14.1) nmol / L, P <0.05〕 and decreased the content of MDA in renal tissue 〔(16.55 ± 2.23) nmol / L, P <0.05]. There was no significant difference in water content between the three groups of rabbits kidney tissue. Conclusion Morphine hypoxic preconditioning may reduce the production of MDA in kidney and increase the activity of SOD in hypoxia-reoxygenation injury, and thus play a protective role in the kidney of hypoxia-reoxygenation injury in rabbits.
