本文应用茚三酮与氨基酸的显色反应 ,研究了差示分光光度法测定不同含量氨基酸的方法。在 p H=8.0 4的缓冲溶液中 ,茚三酮与氨基酸生成蓝紫色络合物 ,最大吸收波长λmax=5 70 nm,表观摩尔吸光系数ε=7.6× 1 0 3 L· mol- 1· cm- 1,在 2 5 m L体积中 ,氨基酸含量在 0 .2 0 0 0~ 0 .6 0 0 0 mg(或 0 .5 0 0 0~ 0 .90 0 0 mg)范围内符合比耳定律。该法简便快速 ,准确度高 ,测定范围较广 ,适用于生物样 (毛发、大蒜、红枣等 )、肥料、饲料中常量特别是高含量氨基酸的测定。
In this paper, the color reaction of ninhydrin with amino acids was applied to study the method of differential spectrophotometric determination of amino acids with different contents. In the buffer solution of p H = 8.0 4, the ninhydrin produced a blue-purple complex with amino acids, the maximum absorption wavelength λmax = 5 70 nm, the apparent molar absorptivity ε = 7.6 × 10 3 L · mol -1 · cm-1, the content of amino acids in the volume of 25 m L conformed to the ratio of Bt in the range of 0.200-0.600 mg (or 0.500-0.090.0 mg) law. The method is simple, rapid, accurate and has a wide range of determination. It is suitable for the determination of biological samples (hair, garlic, red dates, etc.), fertilizers and feedstuffs, especially high amino acids.
