目的研究细胞分裂周期蛋白42(cell division cycle42,Cdc42)基因在阿萨希毛孢子菌的菌丝相和酵母相中的表达差异,探讨其在菌丝生长过程中的作用。方法分别培养标准株酵母相和菌丝相组、标准株不同时间生长组、4株不同来源菌株生长组,检测三组阿萨希毛孢子菌菌丝生成率,并提取总RNA,采用荧光定量PCR方法测定各组中Cdc42的基因表达,并计算其相对表达量。结果不同条件下菌丝生成率不同,Cdc42在各组中均有表达,且随着菌丝生成率的升高,Cdc42基因表达量基本呈上升趋势。结论 Cdc42参与菌丝的形成。
Objective To study the difference of cell division cycle42 (Cdc42) gene expression in mycelial and yeast phases of C. albactercens, and to explore its role in mycelial growth. Methods The standard strains of yeast phase and mycelial phase group, standard strains of different growth groups, four strains of different strains of growth were cultured to detect the rate of mycelial growth of Acipenser ceax in three groups, and the total RNA was extracted and quantified by fluorescence quantitative PCR method was used to determine the gene expression of Cdc42 in each group, and calculate the relative expression. Results Under different conditions, the mycelial growth rates were different, and Cdc42 was expressed in all groups. With the increase of mycelial growth rate, Cdc42 gene expression increased. Conclusion Cdc42 is involved in the formation of mycelium.
