目的 了解视网膜下液 (subretinal fluid,SRF)对培养的视网膜色素上皮 (retinal pigmentepithelium,RPE)细胞、成纤维细胞 (fibroblast,FB)内 Bcl- 2蛋白表达的影响。 方法 应用 Western-blotting技术及免疫组化技术分别检测视网膜下液 (subretinal fluid,SRF)作用于 RPE细胞、FB一定时间后细胞内 Bcl- 2的表达水平。 结果 在 SRF作用 4h后两种细胞内 bcl- 2表达量与无血清培养基静息的对照组相比都有不同程度的增加 ;作用 36 h后 ,所有实验组和对照组细胞内 bcl- 2表达量都下降 ,但 SRF作用下的 RPE细胞和 FB内 bcl- 2水平的下降幅度较为明显 ,而对照组在实验观察期末细胞内 bcl- 2蛋白水平仍维持在较高的水平。 结论 实验早期 SRF具有促进 RPE细胞、FB内 Bcl- 2表达的作用 ,但作用一定时间后加速细胞内 bcl- 2蛋白的降解
Objective To investigate the effect of subretinal fluid (SRF) on the expression of Bcl-2 in cultured retinal pigmentepithelium (RPE) cells and fibroblasts (FB). Methods Western-blotting and immunohistochemistry were used to detect the expression of Bcl-2 in subretinal fluid (SRF) on RPE cells after a certain period of time. Results The expression of bcl-2 in the two cells increased with the increase of bcl-2 level after SRF treatment for 4h compared with the control group without serum-free medium. After 36 hours, the expression of bcl-2 However, the decrease of bcl-2 level in RPE cells and FBs under the action of SRF was more obvious than that in control group, while the level of bcl-2 protein in the control group remained at a high level at the end of the experimental period. Conclusion SRF could promote the expression of Bcl-2 in RPE cells and FB in early stage of experiment, but accelerate the degradation of bcl-2 protein after a certain period of time
