目的观察小檗碱对大鼠胃醋酸性溃疡的愈合作用。方法采用改良 Okabei 法,用80%醋酸诱发大鼠慢性胃溃疡模型,随机分2组,对照组(n=6)以生理盐水0.5 ml/d 灌胃,实验组(n=6)给小檗碱0.1 g/(kg·d)灌胃,连续10 d。11 d 后将鼠断颈处死,剖腹取胃,测量溃疡的横径和竖径,求出平均半径,计算溃疡面积(mm~2)。取大鼠(n=8)实验前禁食24 h(自由饮水),实验时先用戊巴比妥(50 mg/kg,ip)麻醉,用灌流方法每过30 min收集1次胃引流液,第1小时收集的为基础胃液,1 h 后经十二指肠注入小檗碱(0.1 g/kg),每隔30min 收集1次,共4次,最后将所收集的胃液分别用0.1 mol/L NaOH 滴定,测量酸度和计算酸排出量。结果溃疡面积由对照组的0.37±0.08 mm~2减少到实验组的0.11±0.02 mm~2(P<0.05)。小檗碱抑制大鼠基础胃酸的分泌,对照组的30 min总酸排出量为0.11±0.05(mmol/30 min),给药后30,60,90及120 min 分别是0.05±0.01,0.04±0.01,0.03±0.01及0.02±0.01(mmol/30min),给药后60,90及120 min 胃酸总排出量明显减少(P<0.01)。结论本实验发现小檗碱对大鼠慢性胃溃疡有促进愈合作用,机制为小檗碱抑制胃酸作用有关。
Objective To observe the healing effect of berberine on gastric ulcer in rats. Methods The model of chronic gastric ulcer induced by 80% acetic acid in rats was induced by modified Okabei method. The rats were randomly divided into 2 groups. The control group (n = 6) was given gavage with saline 0.5 ml / d. The experimental group (n = 6) Base 0.1 g / (kg · d) gavage for 10 days. After 11 days, the mice were sacrificed and the stomach was taken by caesarean section. The transverse diameter and vertical diameter of the ulcer were measured. The average radius was calculated and the ulcer area (mm ~ 2) was calculated. Rats (n = 8) were fasted for 24 hours (free drinking) before experiment. Animals were anesthetized with pentobarbital (50 mg / kg, ip), and gastric perfusate , The first hour based on the collection of gastric juice, 1 h after injection of berberine duodenum (0.1 g / kg), collected once every 30min, a total of 4 times, and finally the collected gastric juice were 0.1 mol / L NaOH titration, acidity measurement and calculation of acid output. Results The ulcer area decreased from 0.37 ± 0.08 mm 2 in the control group to 0.11 ± 0.02 mm 2 in the experimental group (P <0.05). Berberine inhibited the basal gastric acid secretion in rats. The total acid excretion at 30 min in control group was 0.11 ± 0.05 (mmol / 30 min). At 30, 60, 90 and 120 min after administration, berberine was 0.05 ± 0.01, 0.04 ± 0.01, 0.03 ± 0.01 and 0.02 ± 0.01 (mmol / 30min) respectively. The total gastric acid excretion decreased significantly at 60, 90 and 120 min after administration (P <0.01). Conclusion This experiment found that berberine can promote the healing of chronic gastric ulcer in rats, the mechanism of berberine on the inhibition of gastric acid.
