目的:研究天钩降压胶囊中有效成分的大鼠离体小肠吸收动力学。方法:采用外翻肠囊法,用HPLC测定不同取样时间肠内液天麻苷、丹皮酚的含量,计算累计吸收量,以天麻苷、丹皮酚的累计吸收量对时间作线性回归分析,计算吸收动力学参数。结果:天麻苷回归方程为Y=0.000 6X-0.012 8(r=0.993 0),Ka=6.0×10-5μg.min-1.cm-2,t1/2=1.16×104h;丹皮酚回归方程为Y=0.000 68X+0.001 1(r=0.983 5),Ka=6.8×10-5μg.min-1.cm-2,t1/2=1.02×104 h。结论:天钩降压胶囊中天麻苷、丹皮酚在大鼠离体小肠的吸收为一级动力学过程。
OBJECTIVE: To study the kinetics of isolated small intestine absorption in rats of Tian hookjiangya capsule. Methods: The contents of gastrodia glycosides and paeonol in the intestine of different sampling time were determined by HPLC, and the cumulative absorption was calculated. The cumulative absorption of gastrodin and paeonol were linearly analyzed with time, Calculate absorption kinetic parameters. Results: The regression equation of gastrodin was Y = 0.000 6X-0.012 8 (r = 0.993 0), Ka = 6.0 × 10-5μg.min-1.cm-2 and t1 / 2 = 1.16 × 104h. The results are shown as follows: Y = 0.00068X + 0.001 1 (r = 0.983 5), Ka = 6.8x10-5 μg.min-1.cm-2, and t1 / 2 = 1.02x104 h. Conclusion: The absorption of gastrodia glycosides and paeonol in Tian hook hypotensive capsules in isolated rat intestine is a first-order kinetic process.
