目的:研究酒大黄对实验性动脉粥样硬化(AS)兔纤溶酶原激活物抑制物-1(PAI-1)及其基因的调节作用,探讨酒大黄抗AS机制。方法:将健康雄性新西兰兔32只随机分为4组(n=8):空白对照组(N组)、AS模型组(M组)、酒大黄低剂量组(L组)和酒大黄高剂量组(H组),常规高脂饮食构建动脉粥样硬化模型。治疗给药12周,免疫组织化学法检测主动脉组织PAI-1蛋白表达,半定量逆转录聚合酶链反应(RT-PCR)技术检测兔主动脉组织PAI-1 mRNA的表达。结果:与模型组相比,酒大黄低剂量组能显著抑制主动脉组织PAI-1 mRNA以及蛋白的表达(P<0.05或P<0.01);酒大黄高剂量组显著上调主动脉组织PAI-1 mRNA和蛋白的表达(P<0.05或P<0.01)。结论:酒大黄对AS兔PAI-1蛋白及其mRNA表达的调节作用呈现剂量双重性。
Objective: To study the regulation of wine rhubarb on the expression of plasminogen activator inhibitor-1 (PAI-1) and its gene in experimental atherosclerosis (AS) rabbits and to explore the anti-AS mechanism of wine rhubarb. Methods: Thirty-two male New Zealand white rabbits were randomly divided into 4 groups (n = 8): blank control group (N group), AS model group (M group), wine rhubarb low dose group (L group) Group (H group), conventional high fat diet to construct atherosclerosis model. PAI-1 protein expression in aortic tissue was detected by immunohistochemical method and expression of PAI-1 mRNA in aortic tissue was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results: Compared with the model group, the low dose rhubarb group could significantly inhibit the expression of PAI-1 mRNA and protein (P <0.05 or P <0.01); the high dose rhubarb group significantly up-regulated the expression of PAI-1 mRNA and protein expression (P <0.05 or P <0.01). Conclusion: The effect of rhubarb on PAI-1 protein and its mRNA expression in AS rabbits is dose-dual.
