目的:观察还脑益聪方对老龄大鼠学习记忆功能及脑组织β类淀粉样蛋白(β-AP)和P物质(S.P)水平的影响。方法:以Morris水迷宫试验检测老龄大鼠的空间学习记忆能力,将学习记忆减退的40只大鼠用随机表分成4组,每组10只。即模型(记忆减退老龄大鼠)对照组、还脑益聪方大剂量(13.68 g生药/kg)组、小剂量(6.84 g生药/kg)组和阳性药物对照组(盐酸多奈哌齐片,0.45 mg/kg)组,另取学习记忆能力正常老龄大鼠10只为正常对照组,给与等量水,其余灌胃给药8周。观察测定老龄大鼠的空间学习记忆能力和脑组织匀浆测定β-AP和P物质含量,并检测大鼠血清超氧化物岐化酶(SOD)和丙二醛(MDA)的含量,以HE染色观察大鼠脑组织形态学的病理变化。结果:模型组老龄大鼠在水迷宫中的游泳时间、路径长度明显延长。给药8周后,还脑益聪方大、小剂量组大鼠游泳持续时间、路径长度较模型组显著缩短,寻找平台的方式有所改进。和正常老龄大鼠组比较,模型组大鼠MDA含量显著升高,SOD活力显著降低。与模型组比较,给药组大鼠脑组织的β-AP含量下降和P物质水平增加,血清SOD水平显著升高(P<0.01),脑组织形态结构得到改善。结论:还脑益聪方可降低β-AP在老龄大鼠脑组织的沉积,保护脑组织海马形态的完整性,从而改善老龄大鼠空间学习记忆能力。
Objective: To observe the effect of Huannao Yicong Recipe on the learning and memory function and β-amyloid (AP) and substance P (S) levels in brain of aged rats. Methods: The Morris water maze test was used to detect the spatial learning and memory abilities of aged rats. Forty rats with learning and memory impairment were randomly divided into 4 groups with 10 in each. (Control group), Huannai Yicong large dose (13.68 g crude drug / kg) group, low dose (6.84 g crude drug / kg) group and positive drug control group (donepezil hydrochloride tablets, 0.45 mg / kg) group, and another 10 rats with normal learning and memory ability were used as normal control group and given the same amount of water, while the rest were given gavage for 8 weeks. The contents of β-AP and substance P in brain tissue of rats were measured and the content of superoxide dismutase (SOD) and malondialdehyde (MDA) Pathological changes of rat brain histomorphology were observed by staining. Results: The swimming time and the path length of the aged rats in the model group were significantly longer than those in the water maze. After 8 weeks of administration, the duration of swimming and the length of pathways of rats treated with Huannao Yifeng large and small dose groups were significantly shorter than those of the model group, and the way of looking for the platform was improved. Compared with the normal aging rats, the content of MDA in the model group increased significantly and the activity of SOD decreased significantly. Compared with the model group, the content of β-AP and the level of substance P in the brain tissue of the treated group were increased, and the level of serum SOD was significantly increased (P <0.01). The morphological structure of the brain tissue was improved. Conclusion: Huannao Yicong Decoction can reduce the deposition of β-AP in brain tissue of aged rats and protect the integrity of hippocampal formation in brain tissue, thus improving the spatial learning and memory ability of aged rats.
