目的 探讨黑素转铁蛋白 p97在人脑胶质瘤细胞铁摄取中的作用。 方法 应用含放射性柠檬酸铁(59Fe -Citrate)的培液孵育人脑胶质瘤细胞株GBM及BT32 5 ,以不同浓度PI -PLC及 pronase处理细胞 ,γ -闪烁计数仪测定cpm值 ,得到胶质瘤细胞的相对Fe摄取水平。结果 在一定浓度PI -PLC作用下 ,肿瘤细胞59Fe摄取水平显著降低 ,随着PI -PLC浓度的增加 ,加入 pronase和不加入pronase的细胞59Fe摄取量接近一致。结论 p97在人脑胶质瘤细胞高表达并在其铁摄取中起重要作用
Objective To investigate the role of melanotransferrin p97 in iron uptake in human glioma cells. Methods Human glioma cell lines GBM and BT32 5 were incubated with radioactive ferric citrate (59Fe -Citrate). Cells were treated with different concentrations of PI-PLC and pronase. The cpm values were measured by γ-scintillation counter The relative Fe uptake level of tumor cells. Results Under the action of certain concentration of PI-PLC, the uptake of 59Fe by tumor cells was significantly decreased. With the increase of PI-PLC concentration, the uptake of 59Fe by pronase and without pronase was similar. Conclusion p97 is highly expressed in human glioma cells and plays an important role in its iron uptake
