目的建立全颞下颌关节骨关节病(TMJOA)实验动物模型。方法选择8只新西兰大白兔,在大白兔的右侧颞下颌关节上腔以及下腔注射注射2%木瓜蛋白酶各0.2 mL,注射两次,3天一次,左侧为对照,注射后第1、3、5周取材,对TMJ进行组织病理学观察。结果注射木瓜蛋白酶早期,颞骨关节面、关节盘、髁状突软骨即出现明显的骨关节病的改变;后期出现关节软骨纤维层剥脱,关节软骨以及关节盘中的裂隙明显,局部颞骨关节面和髁状突出现关节软骨剥脱等损害。结论实验结果表明,颞下颌关节上、下腔注入木瓜蛋白酶是造成全颞下颌关节退行性改变动物模型的有效方法,该方法快速、简单、重复性好。
Objective To establish an experimental animal model of TMJOA (total temporomandibular joint osteoarthrosis). Methods Eight New Zealand white rabbits were selected and injected 0.2 mL papain 2% into the superior temporomandibular joint on the right side of rabbits twice a day for three days. 3, 5 weeks of drawing, the histopathological observation of TMJ. Results At the early stage of papain injection, the temporal articular surface, articular disc and condylar cartilage presented obvious osteoarthrosis changes. In the later period, there was exfoliation of articular cartilage fiber layer, obvious cleft in articular cartilage and articular disc, local temporal articular surface and Condyle appears articular cartilage exfoliation and other damage. Conclusion The experimental results show that injection of papain into temporomandibular joint is an effective method to cause the degenerative changes of the whole temporomandibular joint in animal models. The method is rapid, simple and reproducible.
