目的:研究巯基转移酶(thioltransferase,TTase)在年龄相关性白内障患者晶状体上皮细胞中的表达和活性,探讨白内障的发病机制。方法:利用RT-PCR技术检测白内障组和正常对照组晶状体前囊膜下上皮细胞TTasemRNA,以β-actin为参照标化检测指标含量,并比较各组之间TTase表达的差异。结果:RT-PCR结果显示,白内障组晶状体前囊膜下上皮细胞TTase mRNA表达低于正常对照组;白内障皮质组与后囊膜下组差异无显著性,其余各组间差异有显著性。结论:TTase在年龄相关性白内障晶状体前囊膜下上皮细胞中有表达且比正常人低,不同临床类型的表达不同,存在着发病机制的差异。
Objective: To investigate the expression and activity of thioltransferase (TTase) in lens epithelial cells of patients with age-related cataract and to explore the pathogenesis of cataract. Methods: The mRNA of TTasemRNA in the anterior lens epithelial cells of cataract and normal control group were detected by RT-PCR. The content of TTase mRNA was detected by β-actin, and the difference of TTase expression was compared. Results: RT-PCR results showed that the expression of TTase mRNA in the anterior lens epithelial cells of the cataract group was lower than that of the normal control group. There was no significant difference between the cataract group and the posterior subcapsular group, and the difference between the other groups was significant. Conclusion: TTase is expressed in the anterior lens epithelial cells of age-related cataract and is lower than that in normal people. The expression of TTase is different in different clinical types, and the pathogenesis is different.
