尽管脑间质给药被认为是避开血脑屏障阻碍,实现中枢神经系统给药的一种有效途径,但是经过16年的临床和基础研究后发现,只有恶性胶质母细胞瘤经该途径给药有效.本研究对经脑间质途径给予神经保护药治疗脑缺血进行了探索,并首次提出了经脑间质的简单扩散给药技术.结果发现,对大脑中动脉永久栓塞的大鼠模型预先一次性给予胞二磷胆碱(0.0025g/kg,50mmol/L),栓塞12h后显示,SDD组的梗死体积仅是腹腔给药组的1/6,而SDD组胞二磷胆碱的用量却只有腹腔给药组的1/800.本研究表明,选择合适的给药位点,应用简单扩散可实现药物在靶区的分布,并在较长时间内维持有效浓度.
