目的:观察健脾益气之中药复方四君子汤对慢性应激大鼠下丘脑-垂体-肾上腺轴(HPA轴)的影响。方法:设正常组、模型1组、模型2组、四君子汤组,除正常组外其余各组大鼠用电刺激法制备慢性应激动物模型,观察行为学指标的变化,测定血浆皮质酮(CORT)、下丘脑促肾上腺皮质激素释放激素(CRH)和血浆促肾上腺皮质激素(ACTH)含量,同时检测大鼠下丘脑CRHmRNA和垂体ACTHmRNA含量变化。结果:四君子汤能改善大鼠行为表现,降低大鼠血浆ACTH、下丘脑CRH、血浆CORT的含量,控制下丘脑CRHmRNA和垂体ACTHmRNA的过量表达。结论:健脾益气中药复方可有效调整慢性应激大鼠HPA轴的异常。
OBJECTIVE: To observe the effect of Spleen-Qi-Qi Compound Sijunzi Decoction on the hypothalamus-pituitary-adrenal axis (HPA axis) in chronically stressed rats. Methods: Rats in normal group, model 1, group 2 and Sijunzi decoction group were randomly divided into normal group and treated by electrical stimulation to prepare a chronic stress animal model. Changes in behavioral parameters were observed and plasma corticosterone levels were measured. CORT), hypothalamic corticotrophin-releasing hormone (CRH) and plasma adrenocorticotropic hormone (ACTH) levels were measured. Changes in CRH mRNA and pituitary ACTH mRNA levels in the rat hypothalamus were also measured. Results: Sijunzi Decoction can improve the behavior of rats, reduce the plasma ACTH, CRH, plasma CORT content in the rat, and control the hypothalamus CRHmRNA and pituitary ACTH mRNA overexpression. Conclusion: The recipe of Jianpi Qi can effectively regulate the abnormality of HPA axis in chronic stress rats.
