目的研究Fibulin-5对肺癌发生发展的作用及其机制。方法将Fibulin-5过表达质粒和空白质粒转入肺癌细胞中,Western blot和免疫荧光检测β-catenin在核内的定位;在低表达Fibulin-5的肺癌细胞系中过表达Fibulin-5,同时在高表达Fibulin-5的肺癌细胞系中敲低Fibulin-5,Western blot检测p-ERK、p-GSKβ和β-catenin及其下游蛋白的表达;在肺癌细胞中过表达Fibulin-5,进行裸鼠成瘤实验,观察Fibulin-5对肺癌细胞成瘤能力的影响,免疫组化检测两组肿瘤中p-ERK、p-GSKβ和β-catenin及其下游蛋白的表达;免疫组化检测肺癌临床样本中Fibulin-5和β-catenin的表达情况,并进行相关性分析。结果在肺癌细胞中过表达Fibulin-5后β-catenin的核内定位减少;Fibulin-5可以下调p-ERK/p-GSKβ/β-catenin信号通路;Fibulin-5抑制肺癌细胞成瘤能力;肺癌样品中Fibulin-5和β-catenin的表达成负相关。结论 Fibulin-5通过下调β-catenin抑制肺癌的发生发展。
Objective To study the effect of Fibulin-5 on the development of lung cancer and its mechanism. Methods Fibulin-5 overexpression plasmid and plasmid were transfected into lung cancer cells. The localization of β-catenin in the nucleus was detected by Western blot and immunofluorescence. Fibulin-5 was overexpressed in lung cancer cell lines with low expression of Fibulin-5, meanwhile, Fibulin-5 was knocked down in Fibulin-5-overexpressing lung cancer cell lines and the expression of p-ERK, p-GSKβ and β-catenin and its downstream proteins were detected by Western blot. Fibulin-5 was overexpressed in lung cancer cells, The effect of Fibulin-5 on the tumorigenicity of lung cancer cells was observed. The expression of p-ERK, p-GSKβ, β-catenin and its downstream proteins in both groups were detected by immunohistochemistry. The expression of Fibulin-5 and β-catenin in the samples was analyzed and the correlation analysis was carried out. Results Fibulin-5 could downregulate the expression of p-ERK / p-GSKβ / β-catenin in Fibulin-5 cells, Fibulin-5 could inhibit the tumorigenicity of lung cancer cells, The expression of Fibulin-5 and β-catenin in the samples was negatively correlated. Conclusion Fibulin-5 inhibits the development of lung cancer by down-regulating β-catenin.
