目的观察CyclinD1在食管鳞状细胞癌和癌周正常食管组织的表达,从量化角度阐明其在食管鳞状细胞癌组织中表达的病理学意义及其与CyclinE表达的关系。方法用免疫组化MaxVision法检测CyclinD1、CyclinE在食管鳞状细胞癌和癌周正常食管组织的表达,Image-proplus图像分析软件对其表达强度进行定量分析,并用表达的阳性单位(positiveunit,PU)反映其表达强度。结果 CyclinD1蛋白在食管鳞状细胞癌癌细胞核中表达的PU值高于正常食管组织(P<0.001);CyclinD1蛋白表达的PU值与食管鳞状细胞癌分化程度有关(P<0.001);浸润浆膜层组的CyclinD1蛋白表达的PU值高于无浆膜层浸润组(P=0.037);伴淋巴结转移组的CyclinD1蛋白表达的PU值高于无淋巴结转移组(P=0.012);CyclinD1与CyclinE蛋白在食管鳞状细胞癌中表达呈正相关(r=0.650,P<0.01)。结论 CyclinD1在食管鳞状细胞癌中表达上调,且与生物学特性相关,可作为判断肿瘤患者预后不良的标志之一;CyclinD1和CyclinE可能在食管癌癌变过程中起协同作用。
Objective To observe the expression of CyclinD1 in esophageal squamous cell carcinoma and normal esophageal cancer tissues and to elucidate its pathological significance and its relationship with the expression of CyclinE in esophageal squamous cell carcinoma from a quantitative point of view. Methods The expression of CyclinD1 and CyclinE in esophageal squamous cell carcinoma and normal esophageal cancer tissues were detected by immunohistochemistry MaxVision method. The expression intensity was quantified by Image-proplus image analysis software, and positive cells (PU) Reflect the intensity of their expression. Results The PU value of CyclinD1 protein in esophageal squamous cell carcinoma was higher than that in normal esophagus (P <0.001). The PU value of CyclinD1 protein was correlated with the differentiation of esophageal squamous cell carcinoma (P <0.001) The PU value of CyclinD1 protein in the membranous layer was higher than that in the non-serosal layer infiltrating group (P = 0.037). The PU value of CyclinD1 protein in the group with lymph node metastasis was higher than that in the group without lymph node metastasis (P = 0.012) Protein expression in esophageal squamous cell carcinoma was positively correlated (r = 0.650, P <0.01). Conclusions CyclinD1 is upregulated in esophageal squamous cell carcinoma and is related to biological characteristics. CyclinD1 and CyclinE may play a synergistic role in the carcinogenesis of esophageal cancer.
