目的:探讨参芪扶正注射液对荷瘤动物细胞因子Th1/Th2平衡的调节作用,明确该药对生存期的影响及其可能机制。方法:以荷瘤Wistar大鼠为模型,分别给予参芪扶正注射液及顺铂,并以模型组及正常大鼠为对照。用流式细胞仪检测外周血Th1、Th2及其比值,同时用ELISA法检测外周血中IFN-γ和IL-4的含量。结果:化疗组和参芪扶正注射液组Th1/Th2值均高于模型组,P<0.01;但化疗组仍低于空白对照组,P<0.05。参芪扶正注射液组外周血IL-4较模型组和化疗组明显降低,P<0.01;而IFN-γ则明显提高,P<0.01。参芪扶正注射液组生存期较模型组和化疗组明显延长,P<0.01。结论:参芪扶正注射液可以增加Th1类细胞因子,减少Th2类细胞因子,使Th2向Th1漂移,这可能是该药延长荷瘤大鼠生存期的机制之一。
Objective: To investigate the effect of Shenqi Fuzheng injection on the cytokine Th1/Th2 balance in tumor-bearing animals, and to determine the effect of Shenqi Fuzheng injection on the survival period and its possible mechanism. METHODS: The tumor-bearing Wistar rats were treated with Shenqi Fuzheng Injection and Cisplatin, and the model group and normal rats were used as controls. Peripheral blood Th1, Th2 and their ratios were measured by flow cytometry, and IFN-γ and IL-4 levels in peripheral blood were measured by ELISA. Results: The Th1/Th2 values in the chemotherapy group and Shenqi Fuzheng Injection group were higher than those in the model group, P<0.01, but the chemotherapy group was still lower than the blank control group, P<0.05. Shenqi Fuzheng Injection group peripheral blood IL-4 was significantly lower than the model group and chemotherapy group, P <0.01; while IFN-γ was significantly increased, P <0.01. The survival time of Shenqi Fuzheng Injection group was significantly longer than that of model group and chemotherapy group, P<0.01. Conclusion: Shenqi Fuzheng Injection can increase Th1 cytokines, reduce Th2 cytokines, and shift Th2 to Th1, which may be one of the mechanisms of the drug’s prolongation of survival time in tumor-bearing rats.
