目的:观察光敏剂5-氨基酮戊酸(ALA)口服后在肝和胰腺中的代谢,以获得理想的ALA剂量、荧光诊断及治疗时间。方法:给昆明小鼠口服不同浓度的ALA(20mg/kg、40mg/kg、60mg/kg、80mg/kg 4个组别)后,在不同时间点(1小时、2小时、4小时、6小时、8小时、10小时、12小时、24小时、48小时),以波长410nm半导体激光诱导荧光,并通过OMA荧光光谱仪分析检测组织荧光强度,了解ALA不同时间在肝脏、胰腺中的代谢状态。结果:荧光强度在口服ALA40mg/kg组后1小时最强。结论:口服ALA40mg/kg为最适诊断剂量,最佳检测时间为口服ALA40mg/kg后1小时。
OBJECTIVE: To observe the metabolism of photosensitizer 5-aminolevulinic acid (ALA) in the liver and pancreas after oral administration in order to obtain the ideal dose of ALA, fluorescence diagnosis and treatment time. Methods: Kunming mice were treated with different concentrations of ALA (20 mg / kg, 40 mg / kg, 60 mg / kg and 80 mg / kg) for 4 hours and 6 hours at different time points (1 hour, 2 hours, , 8 hours, 10 hours, 12 hours, 24 hours and 48 hours). Fluorescence was induced by semiconductor laser with wavelength of 410 nm. The fluorescence intensity of tissue was detected by OMA fluorescence spectrometer to understand the metabolic state of ALA in liver and pancreas at different times. RESULTS: Fluorescence intensity was the strongest at 1 hour after oral administration of ALA at 40 mg / kg. Conclusion: Oral ALA 40mg / kg is the best diagnostic dose, the best detection time is 1 hour after oral ALA 40mg / kg.
