目的观察他莫昔芬对裸鼠移植性人肝癌细胞生长的影响及应用他莫昔芬后裸鼠肿瘤组织内雌激素受体指标的变化,为他莫昔芬用于肝癌治疗提供实验依据。方法BALB/cA-nu雄性裸鼠26只,背部皮下接种HepG2人肝癌细胞建立裸鼠肝癌模型,14天后成瘤裸鼠随机分成对照组和他莫昔芬治疗组;给药30天后检测各组移植瘤的体积和重量;病理学常规检查,并采用免疫组化检测肿瘤组织内雌激素受体的表达。结果与对照组相比,他莫昔芬治疗组裸鼠肿瘤的体积、重量明显减少(P<0.05),肿瘤内雌激素受体的表达明显减少。结论他莫昔芬通过与肝癌细胞膜上的雌激素受体结合,阻止雌激素作用的发挥,从而抑制肝癌细胞的增殖。
Objective To observe the effect of tamoxifen on the growth of human hepatocellular carcinoma (HCC) in nude mice and the change of estrogen receptor (ER) in the tumor tissue of nude mice after tamoxifen treatment, so as to provide an experimental basis for the treatment of liver cancer with tamoxifen. Methods Twenty-six male BALB / cA-nu nude mice were inoculated subcutaneously with HepG2 human hepatoma cells on the back to establish a model of hepatocellular carcinoma in nude mice. After 14 days, the nude mice were randomly divided into control group and tamoxifen treatment group. The size and weight of the xenograft tumor were observed. Pathological examination was performed routinely. The expression of estrogen receptor in tumor tissue was detected by immunohistochemistry. Results Compared with the control group, the tumor volume and weight of the tamoxifen treated group decreased significantly (P <0.05), and the expression of estrogen receptor in the tumor decreased significantly. Conclusion Tamoxifen inhibits the proliferation of hepatocellular carcinoma cells by inhibiting the action of estrogen by binding with the estrogen receptor on the membrane of hepatocellular carcinoma cells.
