目的:建立测定人血浆丙泊酚(异丙酚) 浓度的HPLC 方法,为临床异丙酚的药理学研究提供科学的分析技术。方法:用环己烷提取血浆中的异丙酚,用HPLC测定异丙酚含量。测定异丙酚以μBondapak C18 柱为分离柱,乙腈水(55∶45)为流动相,以百里酚为内标物,用紫外检测,波长270 nm 。结果:本方法回收率高,重现性好,线性范围为0-1 ~4-0 μg·ml- 1 ,最低检测浓度为0-05 μg·ml- 1 。结论:本方法灵敏、快速、选择性强,具有较高的精密度和准确度,适用于临床血药浓度检测。
OBJECTIVE: To establish a HPLC method for the determination of propofol in human plasma and to provide a scientific analytical technique for the pharmacological study of propofol in clinical practice. Methods: Propofol in plasma was extracted with cyclohexane and propofol content was determined by HPLC. Determination of propofol to μ Bondapak C18 column as the separation column, acetonitrile water (55:45) as the mobile phase to thymol as an internal standard, with UV detection wavelength of 270 nm. Results: The method has high recovery and good reproducibility. The linear range is 0-1 ~ 4-0 μg · ml-1, and the lowest detection concentration is 0-05 μg · ml-1. Conclusion: The method is sensitive, rapid, selective, with high precision and accuracy, suitable for clinical blood concentration detection.
