目的 :探讨 P5 3、P2 1、cyclin D1 、CDk4基因及蛋白在食管癌发生中的意义及其相互间的作用。方法 :采用 L SAB免疫组化染色法 ,对 43例食管癌手术标本的 P5 3、P2 1、cyclin D1 、CDk4蛋白进行标记。结果 :在鳞癌组织中 ,四种抗体的阳性物质存在于细胞核中和或胞浆中 ,在正常鳞状上皮主要存在于核中 ,且从正常鳞状上皮不典型增生鳞癌组织 ,P5 3的阳性表达逐渐上升 ,P2 1的阳性表达却随之下降 ;cyclin D1 、CDk4的阳性率则随 P2 1的下降而呈上升趋势。统计学处理 ,正常上皮与癌组织之间四种抗体的阳性率有显著差异性 (P值均小于 0 .0 5 ) ;同时 ,随着癌分化程度下降 ,P5 3、P2 1的阳性率在 级与 级鳞癌之间差异有显著性 (P<0 .0 5 ) ,但未检出 cyclin D1 、CDk4与癌分化的关系。结论 :p5 3、p2 1、cyclin D1 、CDk4蛋白表达的变化是食管癌发生中的早期事件 ,其构成的调节通路障碍可能参与了食管鳞癌的发生。
Objective: To explore the significance of P5 3, P21, cyclin D1, CDk4 genes and proteins in the development of esophageal cancer and their interactions. METHODS: P53, P21, cyclin D1, and CDk4 proteins from 43 surgical specimens of esophageal cancer were labeled with L SAB immunohistochemical staining. RESULTS: In squamous cell carcinoma tissues, the positive substances of the four antibodies exist in the nucleus and in the cytoplasm, and in the normal squamous epithelium mainly in the nucleus, and from the normal squamous epithelial dysplasia squamous cell carcinoma tissue, P5 3 The positive expression of P21 increased gradually, but the positive expression of P2 1 decreased. The positive rate of cyclin D1 and CDk4 increased with the decrease of P2 1. Statistical analysis showed that the positive rates of four antibodies between normal epithelium and cancer tissue were significantly different (P values were all less than 0.05). Meanwhile, as the degree of cancer differentiation decreased, the positive rate of P5 3 and P2 1 was There was a significant difference between grade and grade squamous cell carcinoma (P < 0.05), but the relationship between cyclin D1 and CDk4 and carcinoma differentiation was not detected. Conclusion : The changes of p53, p21, cyclin D1 and CDk4 protein expression are the early events in the occurrence of esophageal cancer. The disorder of the regulatory pathway may be involved in the occurrence of esophageal squamous cell carcinoma.
