骨髓细胞低温(4℃)保存的实验研究和临床初步应用

本文通过10只兔和13例人骨髓细胞4℃保存,证明TC199对骨髓细胞4℃保存是有效的。EDTA·Na_2对骨髓细胞的抗凝能力比枸橼酸钠强,肝素抗凝维持时间短,容易产生凝结。TC199-EDTA·Ha_2(TCE)保存液与等量骨髓细胞混悬后,在4℃保存3天,骨髓细胞的台盼蓝和伊红拒染细胞的百分率和有核细胞回收率均在70%以上,骨髓粒系祖细胞(Granulocyte-Macrophage Colony-Forming Unit In Culture,简称CFU-C)的回收率为80%,保存5天时骨髓细胞中的粒细胞将出现固缩等退行性变化。临床资料初步表明,4℃保存3天的TCE骨髓细胞的输注是安全的,未见异常反应,可以加速放疗和化疗后骨髓及血液学的恢复。 In this paper, 10 rabbits and 13 human bone marrow cells were preserved at 4 ℃, which proved that TC199 is effective for the preservation of bone marrow cells at 4 ℃. The percentage of trypan blue and eosin-resistant cells and the recovery rate of nucleated cells in bone marrow cells were both 70% after being stored at 4 ℃ for 3 days after suspension of TC199-EDTA · Ha_2 (TCE) , The recovery rate of granulocyte-Macrophage Colony-Forming Unit In Culture (CFU-C) was 80%, and the granulocytes in myeloid cells would degenerate and degenerate after 5 days of storage. Preliminary clinical data showed that transfusion of TCE bone marrow cells preserved at 4 ℃ for 3 days was safe and no abnormal reaction was found, which could accelerate the recovery of bone marrow and hematology after radiotherapy and chemotherapy.
