目的研究骨髓基质中是否具有血液血管干细胞的特征。方法分离并培养人胎儿骨髓基质细胞(hfMSCs),FACS检测细胞的免疫表型。将此细胞接种在基底膜胶中,由血管内皮细胞生长因子(VEGF)和碱性成纤维细胞生长因子(bFGF)诱导,免疫组化和透射电镜检测血管和造血的分化形成。结果此细胞群体的典型特征是CD34阴性的成纤维样细胞99%表达Flk1(即血管内皮细胞生长因子受体2),并能形成血管样结构,而在管样结构形成的过程中诱导出CD34阳性的圆形细胞。结论Flk1+CD34-的hfMSC能向血管内皮细胞和造血细胞分化,提示其具有血液血管干细胞的特征表型。
Objective To investigate whether bone marrow stroma has the characteristics of hemangiocasal stem cells. Methods Human fetal bone marrow stromal cells (hfMSCs) were isolated and cultured. The immunophenotypes of cells were detected by FACS. The cells were seeded in basement membrane glue and induced by vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF). Immunohistochemistry and transmission electron microscopy were used to detect the differentiation of blood vessels and hematopoiesis. Results A typical feature of this cell population is that 99% of CD34-negative fibroblast-like cells express Flk1 (vascular endothelial growth factor receptor 2) and form vasculature, whereas CD34 is induced during tube-like formation Positive round cells. Conclusion Flk1 + CD34- hfMSC can differentiate into vascular endothelial cells and hematopoietic cells, suggesting that it has the characteristic phenotype of hemangiocasal stem cells.
