目的:筛选肾透明细胞癌组织差异表达的miRNAs,并分析其与可能的靶基因作用调控关系。方法:收集8例新鲜的肾透明细胞癌及其癌旁正常肾组织,抽提总RNA,并行Hy3荧光标记,将标记的小RNA与包含1891条探针的miRNA芯片进行杂交,再扫描及数据分析处理。miRNA芯片结果的可靠性采用RT-PCR方法验证。结果:肾透明细胞癌组织与癌旁正常肾组织比较,存在较多明显差异表达的miRNAs,其中上调miRNAs 106个、下调miRNAs 184个,其中111个miRNAs为16.0版miRNA芯片新增的;显著上调的miRNAs 3个、下调的miRNAs 38个。结论:肾透明细胞癌组织的miRNA表达谱有较强特异性,其与可能的靶基因调控关系复杂,其中miR-155、miR-141、miR-200c及let-7家族等与肾透明细胞癌的发病密切相关。
Objective: To screen miRNAs differentially expressed in renal clear cell carcinoma tissues and to analyze their relationship with possible target gene regulation. Methods: Eight fresh renal clear cell carcinomas and their adjacent normal renal tissues were collected. Total RNA was extracted and hybridized with Hy3 fluorescent tag. The labeled small RNA was hybridized with 1891 probe miRNA microarray. Analysis and processing. The reliability of miRNA chip results was verified by RT-PCR. Results: Compared with adjacent normal renal tissues, the number of clear differentially expressed miRNAs in renal clear cell carcinoma tissues was significantly higher than that in adjacent normal tissues (106 of them were up-regulated and 184 of them were down-regulated), of which 111 miRNAs were significantly increased 3 miRNAs and 38 down-regulated miRNAs. CONCLUSIONS: The miRNA expression profile of renal clear cell carcinoma is highly specific and has a complex relationship with the possible regulation of target genes. Among them, miR-155, miR-141, miR-200c and let-7 family are associated with clear cell renal cell carcinoma The incidence is closely related.
