目的:观察粉防己碱对自发性高血压大鼠心肌胶原和心肌僵硬度的作用。方法:测定羟脯氨酸表示胶原的量,胃酶抽提法和SDS-PAGE方法测定胶原Ⅰ/Ⅲ型比例,离体灌注大鼠心脏测定左室心肌舒张僵硬度。结果:粉防己碱显著降低心肌胶原浓度和含量,降低胶原Ⅰ/Ⅲ型比例,改善心肌舒张僵硬度。结论:粉防己碱因其有效的抗纤维化作用,能逆转心肌舒张功能障碍。
Objective: To observe the effect of tetrandrine on myocardial collagen and myocardial stiffness in spontaneously hypertensive rats. Methods: Hydroxyproline was used to determine the amount of collagen. Gastric enzyme extraction and SDS-PAGE were used to determine the ratio of collagen type Ⅰ / Ⅲ. The left ventricular diastolic stiffness was measured by perfusion in rat heart. Results: Tetrandrine significantly decreased the concentration and content of myocardial collagen, reduced the ratio of type Ⅰ / Ⅲ collagen, and improved the diastolic stiffness of myocardium. Conclusion: Tetrandrine can reverse myocardial diastolic dysfunction due to its effective anti-fibrosis effect.
