[目的]探讨光照对扯根菜生长发育的影响。[方法]采用组织培养法获取扯根菜组培苗和黄化苗。每隔6天分别取样,共6次。研究扯根菜黄化苗转绿过程中茎段内脯氨酸、叶绿素、可溶性蛋白含量以及POD活性的动态变化。可溶性蛋白含量测定采用考马斯亮蓝G-250染色法;POD活性测定采用愈创木酚比色法;游离脯氨酸含量测定采用3%磺基水杨酸法;叶绿素测定采用乙醇提取法。[结果]黄化苗的叶绿素和可溶性蛋白含量以及POD活性均低于正常苗,而脯氨酸含量高于正常苗;黄化苗见光后的30d内,叶绿素含量和POD活性一直处于上升趋势,并最终与正常苗相近;可溶性蛋白与脯氨酸的含量总体呈现先升后降的趋势,最终也接近正常苗;表明黄化苗的光形态建成基本完成。
[Objective] The research aimed to discuss the influence of light on the growth and development of pull root vegetables. [Method] Tissue culture seedlings and yellow seedlings were obtained by tissue culture method. Samples were taken every 6 days for a total of 6 times. The dynamic changes of proline, chlorophyll, soluble protein content and POD activity in the stem segments of P. Soluble protein content was determined by Coomassie brilliant blue G-250 staining; POD activity was determined by guaiacol colorimetry; free proline content was determined by 3% sulfosalicylic acid method; chlorophyll determination by ethanol extraction. [Result] The contents of chlorophyll, soluble protein and POD in etiolated seedlings were lower than those in normal seedling, while the content of proline in seedlings was higher than that in normal seedlings. The chlorophyll content and POD activity of yellow seedlings were all on the rise within 30 days , And eventually with the normal seedlings similar; soluble protein and proline content showed an upward trend first and then decreased, and ultimately close to the normal seedlings; that light of the yellow seedlings light form basically completed.
