目的观察BTW5对体外培养的日本血吸虫成虫和童虫的杀灭效果。方法小鼠感染日本血吸虫尾蚴18 d后用肝门静脉灌注法收集童虫,感染5周后用同法收集雌、雄成虫,分别体外培养于DMEM培养液中,加入不同浓度BTW5连续培养3 d,观察虫体死亡及活力降低情况,并设空白对照组。对虫体进行盐酸卡红染色,观察其损伤情况。结果在含不同浓度BTW5的DMEM培养液中,日本血吸虫成虫及童虫的死亡率与活力降低率均显著高于对照组(P均<0.05)。盐酸卡红染色直观显示BTW5对雌雄虫的体表和肠管有损伤作用,另外对雌虫的卵巢也有损伤。结论BTW5具有体外杀灭日本血吸虫成虫和童虫的作用。
Objective To observe the killing effect of BTW5 on adult Schistosoma japonicum and Schistosoma japonicum cultured in vitro. Methods The mice were infected with Schistosoma japonicum cercariae for 18 days and collected by portal vein perfusion method. After 5 weeks of infection, female and male adults were collected and cultured in DMEM culture medium in vitro. The cells were cultured in different concentrations of BTW5 for 3 days. Observe the death and vitality of parasites, and set a blank control group. On the parasites hydrochloric acid card red staining, observe the damage. Results The mortality and viability of Schistosoma japonicum adult and Schistosoma japonicum in DMEM containing different concentrations of BTW5 were significantly higher than those in control group (all P <0.05). Hydrochloric acid card red staining visualization of BTW5 on the male and female body surface and bowel injury, in addition to the female ovary damage. Conclusion BTW5 has the effect of killing Schistosoma japonicum adults and schistosomula in vitro.
