[目的]探讨白藜芦醇诱导食管癌Eca109细胞凋亡的作用机制。[方法]MTT比色法测定细胞增殖的抑制率;透射电镜下观察细胞凋亡形态学变化;RT-PCR法和流式细胞术(FCM)检测白藜芦醇对Eca109细胞survivin和bax表达的影响。[结果]白藜芦醇对Eca109细胞生长有抑制作用,抑制率达76.42%,并呈剂量—时间效应关系。白藜芦醇处理细胞后可见凋亡形态学变化,部分细胞体积变小,染色质浓缩及边集。RT-PCR法和FCM检测结果显示白藜芦醇可抑制survivin上调,促进bax的表达。[结论]白藜芦醇能诱导食管癌Eca109细胞凋亡,其机制可能与调节survivin和bax的表达有关。
[Objective] To investigate the mechanism of resveratrol inducing apoptosis of esophageal cancer Eca109 cells. [Methods] MTT assay was used to determine the inhibition of cell proliferation. Morphological changes of apoptosis were observed under transmission electron microscope. The expression of survivin and bax of resveratrol on Eca109 cells was detected by RT-PCR and flow cytometry (FCM) influences. [Result] Resveratrol had an inhibitory effect on the growth of Eca109 cells, with a inhibition rate of 76.42% and a dose-time effect. Resveratrol treatment of cells showed apoptotic morphological changes, part of the smaller cells, chromatin condensation and edge sets. RT-PCR and FCM results showed that resveratrol could inhibit the up-regulation of survivin and promote the expression of bax. [Conclusion] Resveratrol can induce the apoptosis of esophageal cancer Eca109 cells, which may be related to the regulation of the expression of survivin and bax.
