目的探讨姜黄素对博莱霉素诱导肺纤维化大鼠肺组织Bcl-xl基因表达的影响。方法选取SD大鼠96只,随机分为空白组、模型组、醋酸泼尼松龙组、姜黄素组,每组各24只。各组动物于7、14、28 d随机处死8只,取肺组织采用RT-PCR检测Bcl-xl mRNA的表达和免疫组化法检测蛋白的表达,观察姜黄素对博莱霉素诱导肺纤维化大鼠肺组织Bcl-xl基因表达的影响。结果模型对照组Bcl-xl蛋白与Bcl-xl mRNA表达在第7天开始下降,第14天达最低峰,第28天略有回升,但仍低于空白对照组,姜黄素组Bcl-xl mRNA及蛋白表达显著高于模型对照组及空白对照组,均有统计学意义(P<0.01);姜黄素组与泼尼松龙组比较7、14 d均无统计学意义(P>0.05),28 d姜黄素组Bcl-xl表达高于泼尼松龙组,有统计学意义(P<0.01)。结论在博来霉素诱导肺纤维化形成阶段应用,姜黄素干预可能通过上调Bcl-xl mRNA及蛋白的表达减轻肺纤维化的程度。
Objective To investigate the effect of curcumin on the expression of Bcl-xl gene in lung tissue of bleomycin-induced pulmonary fibrosis in rats. Methods Ninety-six SD rats were randomly divided into blank group, model group, prednisolone acetate group and curcumin group, 24 in each group. Animals were sacrificed on the 7th, 14th, and 28th day for 8 animals. The expression of Bcl-xl mRNA was detected by RT-PCR and protein expression was detected by immunohistochemistry. The curcumin-induced bleomycin-induced pulmonary fibers were observed. Effect of Bcl - xl Gene Expression in Rat Lungs . Results The expression of Bcl-xl protein and Bcl-xl mRNA in the model control group began to decline on the 7th day, reached the lowest on the 14th day, and slightly increased on the 28th day, but was still lower than the blank control group. The Bcl-xl mRNA in curcumin group The protein expression was significantly higher than that of the model control group and the blank control group (P<0.01). There was no significant difference between the curcumin group and the prednisolone group on the 7th and 14th day (P>0.05). The expression of Bcl-xl in curcumin group was higher than that in prednisolone group at 28 days (P<0.01). Conclusion In the application of bleomycin-induced pulmonary fibrosis, curcumin intervention may reduce the degree of pulmonary fibrosis by up-regulating the expression of Bcl-xl mRNA and protein.
