【目的】探讨慢性增强型体外反搏对高胆固醇血症猪主动脉血管内皮细胞凋亡的影响。【方法】18头雄性乳猪随机分为正常饲养组(n=6),高脂饲养组(n=6)及高脂饲养+反搏组(n=6)。后2组复制高胆固醇血症猪模型并对高脂饲养+反搏组进行为时36h的慢性增强型体外反搏,收集3组动物主动脉血管内皮细胞,TUNEL法测定其主动脉血管内皮细胞的凋亡指数。【结果】与正常饲养组相比,高脂饲养组和高脂饲养+反搏组血清总胆固醇和低密度脂蛋白明显升高。主动脉血管内皮细胞凋亡指数在正常饲养组为(127±36)‰,在高脂饲养组为(237±23)‰,在高脂饲养+反搏组为(177±12)‰,高脂饲养组和高脂饲养+反搏组较正常饲养组明显升高(P<0.05),高脂饲养+反搏组较高脂饲养组明显降低(P<0.05)。【结论】高胆固醇血症刺激血管内皮细胞凋亡,慢性增强型体外反搏通过减轻血管内皮细胞凋亡拮抗高胆固醇脂血症对血管内皮的损伤作用。
【Objective】 To explore the effect of chronic enhanced extrinsic counterpulsation on apoptosis of porcine aorta vascular endothelial cells in hypercholesterolemia. 【Methods】 Eighteen male piglets were randomly divided into normal feeding group (n = 6), high fat feeding group (n = 6) and high fat feeding + counterpulsation group (n = 6). The latter two groups were hypercholesterolemic pig model and chronic high-fat external counterpulsation was performed on the high-fat diet + counterpulsation group for 36 hours. Three groups of animal aortic endothelial cells were collected. TUNEL assay was used to determine the aortic endothelial cells The apoptosis index. 【Result】 Compared with normal feeding group, serum total cholesterol and low density lipoprotein were significantly increased in high fat diet group and high fat diet + EECP group. The apoptosis index of aortic endothelial cells was (127 ± 36) ‰ in normal feeding group, (237 ± 23) ‰ in high fat feeding group and (177 ± 12) ‰ in high fat feeding + counterpulsation group The lipid diet group and the high-fat diet + EECP group were significantly higher than those of the normal diet group (P <0.05), while those in the high fat diet group and the EECP group were significantly lower than the high fat diet group (P <0.05). 【Conclusion】 Hypercholesterolemia stimulates the apoptosis of vascular endothelial cells. Chronic enhanced EECP can inhibit the injury of vascular endothelial cells by reducing the apoptosis of vascular endothelial cells by hypercholesterolemia.
