目的:检测annexin Ⅰ,Grp94,p53,Ki-67四种侯选肿瘤标志物在食管癌及癌前病变中的表达情况,为食管癌的早期检测提供依据. 方法:72例单纯手术切除食管鳞癌及其癌旁伴有的不同程度癌前病变,构建组织芯片;应用免疫组化方法检测annexin Ⅰ,Grp94,突变型p53,Ki-67蛋白在食管鳞癌及不同程度癌前病变中的表达差异. 结果:四种蛋白在食管癌前病变中均有异常表达,与正常鳞状上皮比较存在显著性差异:annexin Ⅰ在低度癌前病变组(轻度不典型增生/中度不典型增生)丢失率为68.3%,(P=0.000);p53和Ki-67的阳性表达率分别为67.6%和71.1%(P=0.000);Grp94在早期浸润性癌呈极显著过度表达,表达增强率62.0%(P=0.000). 结论:Annexin Ⅰ,Ki-67,p53蛋白为食管癌早期预警研究提供了重要线索;Grp94蛋白作为食管鳞癌进展期标志物具有参考价值.
OBJECTIVE: To detect the expression of four candidate tumor markers annexin Ⅰ, Grp94, p53 and Ki-67 in esophageal cancer and precancerous lesions, and to provide evidence for the early detection of esophageal cancer.Methods: 72 cases of simple surgical resection of esophageal squamous cell carcinoma To detect the expression of annexin Ⅰ, Grp94, mutant p53 and Ki-67 protein in esophageal squamous cell carcinoma and different degrees of precancerous lesions by immunohistochemistry Difference.Results: All four proteins were abnormally expressed in esophageal precancerous lesions, which were significantly different from those in normal squamous epithelium: annexin Ⅰ in low precancerous lesions group (mild dysplasia / moderate dysplasia ) Was 68.3% (P = 0.000). The positive rates of p53 and Ki-67 were 67.6% and 71.1% (P = 0.000) respectively. Grp94 was highly overexpressed in early invasive carcinoma, 62.0%, respectively (P = 0.000) .Conclusion: Annexin Ⅰ, Ki-67 and p53 proteins provide important clues for the early warning of esophageal cancer; Grp94 protein may serve as a reference for the progression of esophageal squamous cell carcinoma.
