目的:观察温度敏感性离子通道TRPV3(TRPV3)在人类膝关节软骨及滑膜组织中的表达,探讨TRPV3在人软骨及滑膜组织中表达的意义。方法收集不同年龄的正常人的膝关节软骨组织标本10例,滑膜组织5例,膝关节骨性关节炎患者的关节软骨、滑膜组织标本各5例,每例软骨组织标本均采用番红“O”染色验证是否发生退行性改变。利用免疫荧光,免疫组化, RT-PCR和Western blot方法检测TRPV3通道在正常人软骨、滑膜和OA软骨、滑膜组织中的表达分布。结果显微镜下骨关节炎组的番红“O”染色显示没有发生退行性改变;免疫荧光,免疫组化,RT-PCR与Western blot方法在软骨及滑膜组织中均检测到TRPV3通道蛋白,且结果显示骨关节炎组的TRPV3的表达量高于正常对照组。Western-blot结果显示随着年龄的增长,TRPV3的表达呈增长趋势,t检验证明有统计学意义(P<0.05)。免疫荧光染色可见TRPV3蛋白均匀的分布在细胞膜上。结论 TRPV3表达增高可能与关节软骨损伤密切相关,可能为今后关节软骨损伤的治疗提供新的靶点。“,”ObjectiveObservation of temperature-sensitive ion channels TRPV3 of the knee cartilage and synovial tissue in humans, to explore the meaning TRPV3 in human cartilage and synovial tissue expression.MethodsCollection of different ages normal knee cartilage tissues 10 cases, five cases of synovial tissue, patients with osteoarthritis of the knee articular cartilage, synovial tissue samples each of five cases, each case of cartilage tissue samples are used safranin “O ”staining verify degenerative changes. Use of immunofluorescence, immunohistochemistry, RT-PCR and Western-blot detection of TRPV3 channel expression profiles in normal human cartilage, synovial and OA cartilage, synovial tissue.ResultsUnder the microscope osteoarthritis group safranin “O” staining showed no degenerative changes; immunofluorescence, immunohistochemistry, RT-PCR and Western-blot method in cartilage and synovial tissue were detected TRPV3 channel protein, and the results Show TRPV3 expression of osteoarthritis group was higher than the normal control group.Western-blot shows that with age, the expression of TRPV3 is a growing trend,t test proved statistically significant (P<0.05). Immunofluorescence staining evenly distributed TRPV3 protein in the cell membrane.ConclusionTRPV3 increased expression may be related to articular cartilage damage, may provide new targets for future treatment of articular cartilage injury.
