目的 检测皮肤癌 p5 3基因 5~ 8外显子的突变频率 ,比较变性梯度凝胶电泳 (DGGE)和单链构象多态性分析 (SSCP)检测p5 3基因突变的敏感性。方法 分别采用DGGE和SSCP ,对 40例皮肤癌患者手术切除组织的 p5 3基因 5~ 8外显子进行突变检测。 结果 皮肤癌 p5 3基因 5~ 8外显子的总突变率为 35 % ,其中鳞状细胞癌 (SCC)的突变率为 36 % ,基底细胞癌 (BCC)为 33%。采用DGGE和SSCP检测的突变率分别为 33%和 2 5 %。结论 35 %的皮肤癌组织存在 p5 3基因 5~ 8外显子突变。DGGE检测p5 3基因突变的敏感性高于SSCP。DGGE结合SSCP有助于提高突变检出率
Objective To detect the mutation frequencies of p53 gene exon 5-8 in skin cancer and compare the sensitivity of p53 gene mutations by denaturing gradient gel electrophoresis (DGGE) and single strand conformation polymorphism analysis (SSCP). Methods DGGE and SSCP were used to detect mutations in the p53 gene 5-8 exon in 40 cases of skin cancer. Results The total mutation rate of 5-8 exons of p53 gene in skin cancer was 35%. The mutation rate of squamous cell carcinoma (SCC) was 36%, and that of basal cell carcinoma (BCC) was 33%. The mutation rates detected by DGGE and SSCP were 33% and 25%, respectively. Conclusion The p53 gene 5-8 exon mutations are found in 35% of skin cancer tissues. The sensitivity of DGGE for detection of p53 gene mutation was higher than that of SSCP.
