目的:建立葛根中β-谷甾醇的测定方法。方法:采用超高效液相色谱法,色谱柱为Agilent ZORBAX RRHD Eclipse Plus C18(2.1×100mm,1.8μm),流动相为甲醇-0.1%乙酸水(100:4单泵),流速为0.15m L/min,检测波长为206nm,柱温为50℃。结果:β-谷甾醇浓度在0.0186~0.186μg范围内线性关系良好,回归方程为y=1260.5x+1.9164(r=0.9996),平均加样回收率为96.68%,RSD(%)值为2.63。结论:该法专属性强、快速、简便、准确,可以用于葛根中β-谷甾醇的含量测定,为葛根的质量控制提供依据。
Objective: To establish a method for determination of β-sitosterol in Radix Puerariae. Methods: The HPLC method was Agilent ZORBAX RRHD Eclipse Plus C18 (2.1 × 100mm, 1.8μm). The mobile phase was methanol-0.1% acetic acid water (100: 4 single pump) with a flow rate of 0.15m L / min, detection wavelength of 206nm, the column temperature is 50 ℃. Results: The linear range of β-sitosterol concentration ranged from 0.0186 to 0.186 μg. The regression equation was y = 1260.5x + 1.9164 (r = 0.9996). The average recoveries were 96.68% and RSD (%) was 2.63. Conclusion: The method is specific, rapid, simple and accurate. It can be used for determination of β-sitosterol in Pueraria lobata, and provide the basis for the quality control of Pueraria lobata.
