应用荧光法研究了以乙二胺为核的聚酰胺-胺型(PAMAM)树枝状高分子及其端基改性衍生物在水溶液中与牛血清白蛋白的相互作用.结果表明,各类树枝状高分子都能引起牛血清白蛋白荧光猝灭,其程度主要取决于各自末端基团的性质.同时我们还考查了体系pH值和离子强度的变化对各类树枝状高分子与BSA相互作用的影响.此外,圆二色谱结果说明各类树枝状高分子的加入会使BSA结构中α-螺旋的程度降低,从而诱导了BSA的二级结构发生改变.
The interaction between polyamide-amine (PAMAM) dendrimers with ethylenediamine as nucleus and modified derivatives of their terminal groups in bovine serum albumin (BSA) was studied by fluorescence spectroscopy. The results showed that all kinds of branches Fluorescence quenching of bovine serum albumin can be induced by macromolecules, the extent of which depends on the nature of the end groups, and we also examine the changes of the system pH and ionic strength on the interaction of various dendrimers with BSA In addition, the results of circular dichroism showed that the addition of various dendrimers decreased the degree of α-helix in BSA and induced the secondary structure of BSA.
