目的:筛选地骨皮调控H2O2所致人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HU-VEC)凋亡的有效部位,并研究相关的信号转导通路,探讨其保护作用的分子机制。方法:用H2O2构建凋亡模型,运用MTS检测细胞存活率,流式细胞仪检测凋亡率筛选地骨皮有效部位,及Western观察相关信号通路蛋白P-AKT、P-NF-KB表达情况。结果:地骨皮30%,50%,70%醉提取部位均可以减少HUVEC的凋亡,可以明显增加P-AKT的表达,降低P-NF-KB活性,尤其55%醇提地骨皮用量最小效果最佳。结论:地骨皮抑制H2O2所致HUVEC凋亡的主要成分在55%醇提部位,它可以通过AKT、NF-κB信号通路起到抗凋亡的作用。
OBJECTIVE: To screen the effective sites of apoptosis induced by H2O2 in human umbilical vein endothelial cells (HU-VEC) and to study the molecular mechanisms involved in the protective effect of the signal transduction pathway. Methods: Apoptosis model was established by H2O2. MTS was used to detect the cell viability. Flow cytometry was used to detect the effective sites of apoptotic cells. The expressions of P-AKT and P-NF-KB were detected by Western blotting. RESULTS: 30%, 50%, 70% DRG could reduce the apoptosis of HUVEC, significantly increase the expression of P-AKT and decrease the activity of P-NF-KB. The best minimum effect. CONCLUSION: The main component of apoptosis of HUVEC induced by H2O2 in Digupi is 55% alcohol extract, which can play an anti-apoptotic role through AKT and NF-|ÊB signaling pathway.
