目的研究献血者乙型肝炎病毒表面抗原118位氮基酸变异对ELISA检测的影响。方法PCR扩增献血者表面抗原基因,恢复变异点氨基酸为野生型,构建不同的表达载体,在293T细胞株中表达,检测各自在不同试剂中的反应格局。结果117位点对检测反应格局不产生明显影响,118位点的变异明显影响检测格局,恢复变异点氨基酸明显提高了检出率和灵敏度。结论118位氮基酸变异影响了乙肝表面抗原的筛查,改变了其的免疫反应原性。
Objective To study the effect of mutation of amino acid at position 118 of hepatitis B virus surface antigen on the detection of ELISA in blood donors. Methods The gene of surface antigen of blood donors was amplified by PCR, and the amino acids at the mutation site were recovered as wild type. Different expression vectors were constructed and expressed in 293T cell line. The reaction patterns in different reagents were detected. Results No significant effect of 117 sites on the reaction pattern was detected. The mutation of 118 locus significantly affected the pattern of detection, and recovery of amino acid at mutation point significantly improved the detection rate and sensitivity. Conclusion The mutation of 118 amino acids affects the screening of hepatitis B surface antigen and changes its immunogenicity.
