目的采用手性固定相高效液相色谱法拆分异丙肾上腺素对映体。方法以氯甲酸苄酯作为非手性衍生化试剂,与异丙肾上腺素进行衍生化反应,通过Chiral PAKRAD-RH手性色谱柱分离。检测波长选择为226 nm,流动相为甲醇-水-四氢呋喃(150∶20∶7),流速0.5 m L·min-1,柱温30℃。结果最大分离度达到1.61,成功折分了异丙肾上腺素对映体。结论所用方法具有操作简单、反应条件温和、分离度高等特点,可为此类药物对映体的工业化分离和制备提供新的方法。
OBJECTIVE To separate enantiomer of isoproterenol by high performance liquid chromatography with chiral stationary phase. Methods Benzyl chloroformate was used as achiral derivatization reagent, derivatized with isoproterenol and separated by Chiral PAKRAD-RH chiral column. The detection wavelength was selected to be 226 nm. The mobile phase was methanol-water-tetrahydrofuran (150:20:7) and the flow rate was 0.5 m L · min-1. The column temperature was 30 ℃. The maximum resolution of 1.61 was obtained and the isoproterenol was successfully separated. Conclusion The method has the advantages of simple operation, mild reaction conditions and high resolution, which can provide a new method for the industrial separation and preparation of enantiomers of these drugs.
