目的研究模拟失重效应条件下大鼠肌组织机械生长因子(mechano-growth factor,MGF)的表达变化,探讨MGF在失重性肌萎缩中的作用。方法采用大鼠尾部悬吊模型模拟失重效应,尾吊1、4、7、10、14天后,分离并分析肌肉相对重量变化,并通过实时定量PCR分析模拟失重效应对比目鱼肌(soleus,SOL)与趾长伸肌(extensor digitorum longus,EDL)中胰岛素样生长因子(insulin-like growth factor,IGF-I)两个选择性剪接体MGF、IGF-IEa及M-钙粘附分子(M-cadherin,M-Cad)mRNA表达水平的影响,同时分析IGF-IEa和MGF在两种肌肉中的表达差异。结果尾吊4 d后,SOL相对重量开始显著下降,而EDL相对重量只有轻微下降趋势。尾吊不同时间后,比目鱼肌的MGF、IGF-IEa和M-Cad等基因表达都表现出显著性下降;而在趾长伸肌中,只表现出轻微的下降趋势。结论在尾部悬吊处理条件下,不同肌肉类型MGF的表达变化不同。推测MGF表达的减少与SOL失重性肌萎缩的发生有关。
Objective To investigate the expression of mechanical growth factor (MGF) in rats under simulated weightlessness and to explore the role of MGF in weightless muscle atrophy. Methods The rat tail suspension model was used to simulate the effect of weightlessness. After the tail suspension for 1, 4, 7, 10 and 14 days, the muscle relative weight changes were analyzed. The effects of simulated weightlessness on soleus muscle (SOL) (IGF-I) and MGF (IGF-I) and M-cadherin (M-cadherin) in two extensor digitorum longus (EDL) , M-Cad) mRNA expression levels, at the same time analysis of IGF-IEa and MGF in two muscle differences in expression. After 4 days of tail suspension, the relative weight of SOL began to decline significantly, while the relative weight of EDL showed a slight downward trend. After tail-suspending for a long time, the gene expression of MGF, IGF-IEa and M-Cad in soleus muscles showed a significant decrease; while in extensor digitorum longiflorum, only a slight downward trend showed. Conclusion Under the condition of tail suspension, the expression of MGF in different muscle types varies. It is speculated that the decrease of MGF expression is related to the occurrence of SOL weightless muscular atrophy.
