Objective To compare the amount of dopamine (DA) uptake in peripheral blood lymphocytes between patients with Parkinson’s disease (PD) and normal controls. Methods Peripheral blood lymphocytes were isolated by density gradient centrifugation. The dopamine transporter (DAT) protein in lymphocytes was detected by Western blot. The uptake of 3H-DA by lymphocytes was measured by liquid scintillation counting. Results Western blot showed that a region corresponding to the size of DAT protein appeared at a molecular weight of 660 0 0. Lymphocytes had the function of uptake of 3H-DA in time, concentration and Na + -dependent manner. The uptake of lymphocytes by PD patients for 3H -DA was significantly lower than the normal control group (P <0.05). Conclusion Detecting PD patients with peripheral blood lymphocyte uptake of 3H-DA and the difference between the normal control group may become a laboratory diagnostic basis for PD
