目的探讨人肿瘤坏死因子α(TNF-α)联合紫杉醇对人黏液表皮样癌细胞系(MEC-1)的抑制增殖作用及与细胞凋亡的关系,为化疗与生物治疗联合应用治疗粘液表皮样癌提供实验依据。方法将人黏液表皮样癌细胞系(MEC-1)和对照组原代培养鼠肺成纤维细胞(LFB)分为9组,每组给予不用药物浓度作用72h。3H-TDR法检测细胞增殖抑制率;流式细胞术检测细胞凋亡率及细胞周期改变;透射电镜观察凋亡细胞形态变化。结果联合用药组的细胞增殖抑制率显著高于单独用药组(P<0.05)。LFB细胞各组的细胞增殖抑制率显著低于MEC-1细胞(P<0.05)。结论TNF-α联合紫杉醇对MEC-1细胞增殖的抑制作用以及诱导凋亡具有协同作用。
Objective To investigate the relationship between tumor necrosis factor-α (TNF-α) and paclitaxel on the proliferation and apoptosis of human mucoepidermoid carcinoma cell line MEC-1 and the relationship between apoptosis and the treatment of mucoepidermoid carcinoma Cancer provides experimental evidence. Methods Human mucoepidermoid carcinoma cell line (MEC-1) and control group primary cultured murine lung fibroblasts (LFB) were divided into 9 groups, each group given drug-free concentration for 72 hours. The inhibitory rate of cell proliferation was detected by 3H-TDR assay. The apoptosis rate and cell cycle were detected by flow cytometry. The morphological changes of apoptotic cells were observed by transmission electron microscope. Results The inhibitory rate of cell proliferation in combination group was significantly higher than that in single group (P <0.05). The inhibitory rate of cell proliferation in LFB cells was significantly lower than that in MEC-1 cells (P <0.05). Conclusion TNF-α combined with paclitaxel can inhibit the proliferation of MEC-1 cells and induce apoptosis.
