目的:研究可用于盐酸小檗碱萃取的双水相体系。方法:先用3种双水相体系进行萃取试验,然后选择较好的一种体系进行单因素试验和正交试验。结果:向10 mL 95%乙醇和10 mL 2.2 mol/L硫酸铵溶液混合形成的双水相体系中加入质量分数为53.22%的盐酸小檗碱粗品600 mg,调节体系pH值至4,70℃水浴处理30 min,萃取率可达99.29%;将萃取液集中在一起,40℃下烘干,所得盐酸小檗碱的质量分数为88.43%。结论:该体系是适合于盐酸小檗碱萃取的双水相体系。
Objective: To study the aqueous two-phase system that can be used for the extraction of berberine hydrochloride. Methods: Firstly, three kinds of aqueous two-phase system were used for the extraction test. Then, a better system was selected for single factor test and orthogonal test. Results: The crude product of berberine hydrochloride with the mass fraction of 53.22% (600 mg) was added into the aqueous two-phase system composed of 10 mL of 95% ethanol and 10 mL of 2.2 mol / L ammonium sulfate solution. The pH of the system was adjusted to 4,70 ℃ Water bath for 30 min, extraction rate of up to 99.29%; the extracts together, drying at 40 ℃, the resulting berberine hydrochloride mass fraction of 88.43%. Conclusion: This system is suitable for the extraction of berberine hydrochloride aqueous two-phase system.
