目的 探讨神经细胞凋亡与缺氧和机械损伤的关系。 方法 体外原代培养大鼠神经元细胞 ,经低氧或机械损伤处理后 ,应用光镜、电镜、末端脱氧核苷酸介导的X -dUTP缺口末端标记 (TdT -mediateddUTPneikendlabeling ,TUNEL)法、流式细胞仪、DNA凝胶电泳及免疫组化分析细胞凋亡及其相关基因表达的改变。 结果 经低氧或机械损伤后神经细胞出现凋亡的形态改变 ;其DNA呈现“梯状”断裂现象 ;细胞凋亡百分率分别为 10 .8%和 4.8% (对照组为 1.6 % ) ,凋亡相关基因 (c-myc、fas、fasL)表达明显增加。结论 缺氧及机械损伤可诱导神经细胞凋亡 ,其分子机制可能与某些凋亡相关基因表达改变有关。
Objective To investigate the relationship between neuronal apoptosis and hypoxia and mechanical injury. Methods Primary cultured rat neuronal cells were exposed to hypoxia or mechanical injury. TdT-mediated terminal deoxynucleotidyl transferase mediated nick end labeling (TUNEL) and flow cytometry Cytometry, DNA gel electrophoresis and immunohistochemistry analysis of apoptosis and related gene expression changes. Results The morphological changes of apoptotic neurons were observed under hypoxia or mechanical injury. The DNA showed “ladder-like” cleavage. The percentages of apoptotic cells were 10.8% and 4.8% (control group, 1.6% Related genes (c-myc, fas, fasL) expression was significantly increased. Conclusion Hypoxia and mechanical injury can induce apoptosis of neural cells, and its molecular mechanism may be related to the changes of some apoptosis-related genes.
